Meaning
now that, since (formal)
〜上は means "now that…, since…" and introduces a resolve or an obligation that naturally follows a decision or fact: now that it is decided, all I can do is move forward. It is the stiff, literary sibling of 〜以上は and 〜からには.
書き言葉 and formal speeches; rare in casual talk, where からには or 以上 is used instead. The second clause expresses determination, duty or a strong judgment (〜べきだ, 〜なければならない, 〜つもりだ, 〜しかない).
